ABSTRACT:
Mutant forms of midivariant RNA are disclosed which are replicated efficiently by the enzyme Q.beta. replicase in the presence of an agent(s) which is useful for the real time, quantitative detection of the accumulating RNA products. This agent(s) also suppresses the replication of non-mutant wild type midivariant RNA. The mutant MDV RNAs, when modified by the addition of specific probe sequences, retain their replication properties in the presence of the detection agent. This method allows the replication to be monitored using a real time mode thereby providing a reliable method for monitoring the kinetics of the reaction, and determining the initial level of amplified RNA.
