ABSTRACT:
The present invention is based on the isolation of a gene coding for streptokinase fromStreptococcus equisimilis(ATCC 9542), its cloning, and expression inE. coli. Two different strategies were carried out for the production of enzymatically-active streptokinase. In the first strategy, streptokinase was expressed as an inclusion body without its signal sequence followed by purification, solubulization, and renaturation to obtain an active preparation. The purified enzyme was formulated and lyophilized. In the second strategy, enzymatically-active streptokinase is secreted into the culture medium. The enzyme was purified, formulated and lyophilized.
