ABSTRACT:
A method for amplifying a nucleic acid fragment is described. A nucleic acid having interspersed repetitive elements is cleaved and the resulting nucleic acid fragments are ligated to a bubble oligonucleotide having two double stranded portions flanking a non-complementary portion consisting of two single strands to make bubble
ucleic acid units. These units are treated with a first primer that is complementary to at least a portion of the interspersed repetitive element, and with a second primer that is complementary to at least a portion of the extension product of the first primer, that portion containing sequences complementary to one of the single strands of the bubble oligonucleotide, under conditions which produce extension products of the primers. Also described are methods for labeling the products of the amplification, methods for detecting chromosomal aberrations and other uses of the amplification products.
