A manufacturing method of a plugged honeycomb structure30which comprises a honeycomb substrate1and plugging portions15arranged so as to form complementary checkered patterns alternately at both end surfaces11and21of the honeycomb substrate1. The method comprises attaching mask films4aand4bto both end surfaces11and21of the honeycomb substrate1, respectively; forming slurry permeation holes3aat portions of the mask films4aand4bcorresponding to cell opening end portions7, respectively; forming deaeration holes3bat portions of the mask films4aand4bfacing the portions provided with the slurry permeation holes3avia cells2; submerging both end surfaces11and21of the honeycomb substrate1to which the mask films4aand4bhave been attached into a plugging slurry6, respectively; and firing the resultant slurry filled substrate20.
